Jackett - who also managed Watford, Millwall and Portsmouth, among other clubs - had been working at the League Two club since a takeover in December 2022.
He leaves his role with the Gills eighth in the table, just one point off the play-off positions.
Jackett joined Wolves in 2013 with the side having been relegated to the third tier of English football, but returned to the Championship at the first attempt as League One champions. He remained in charge until the takeover by Fosun in 2016, when he was sacked shortly after the buyout.
He was replaced by Walter Zenga as head coach at Molineux.
