Clarke was due to play for Heath Hayes with permission from the Rangers boss but the Midland Alliance side had no game on Saturday.
Instead, the 25-year-old turned out for Wolverhampton Casuals against Tividale in the FA Cup without the knowledge of his manager.
Now the wide man cannot play in Rangers' replay at home to Stourport Swifts tomorrow night, after the first game produced a 1-1 draw.
The wide man would have been involved as loanee Luke Bottomer is not allowed to be involved, under the terms of the deal with Tamworth.
And Clowes is set to name seven substitutes after attracting criticism for only taking five to Walshes Meadow for the first game.
He said: "Clarke has cup tied himself without knowing and I am not too pleased, so he's cost himself a place in the squad.
"He would have been involved as we only had five on the bench on Saturday, which was brought up after the game.
"If I take seven, I have got to pay them and I haven't got the abundance of money that previous managers have had.
"It was a negative comment from a fan who also said that we were lucky to still be in the hat, but we should have won.
"When I get asked stupid questions like that, it frustrates me. Every pound that I have got, I have to spend wisely."
