Goalkeeper Martinez and forward Asensio were both absent from Saturday’s 1-0 win at Brentford with muscle pain.
But both have been passed fit to face Brugge in the last-16 second leg, along with midfielder Amadou Onana.
The latter has been missing for more than a month with a calf injury and is now back to bolster Villa’s squad.
Emery’s men hold a 3-1 advantage heading into the match.
“They are both in the squad,” said the Villa boss, when asked about Martinez and Asensio in Tuesday’s pre-match press conference.
“Onana as well is coming back in the squad. Ross Barkley is not, while Donyell Malen and Andres Garcia cannot play.”
The latter pair, both signed during the January transfer window, are not part of Villa’s Champions League squad.
Asensio has scored five goals in six appearances since joining on loan from Paris Saint-Germain in the transfer window, including braces in each of his last two matches at Villa Park.
