Walsall have already secured progression to the next round but can leapfrog Group I leaders Birmingham City with an outright win over bottom-place Shrews.
The Saddlers have collected five points from their opening two games - drawing 1-1 at Birmingham and gaining an extra point in the shoot-out before beating Fulham under-21s 1-0 at Bescot.
Birmingham spearhead the group with seven points and climbed above Walsall into top spot with a 7-1 demolition of Fulham under-21s.
Tommy Simkin, Donervon Daniels, David Okagbue, Liam Gordon and Brandon Comley are all due to jet off on international duty this week and are not expected to be involved.
Harry Williams will also still be unavailable due to concussion along with long-term absentees Priestley Farquharson (thigh) and Josh Gordon (hamstring).
Sadler said: "Naturally I am going to change things around. It's an opportunity to do that.
"We want to win the game. I want to finish top of that group but I also want to make sure that everyone is up to speed and ready for the games when they come along.
"We'll definitely play some of the guys that haven't played recently. That's 100 per cent going to happen but it's with the intention of trying to win the game and making sure everyone is fit and raring to go for the next games."
Walsall are unbeaten in six games across all competitions and have lost just once in 12 games on home soil this season.
Meanwhile, Shrewsbury sunk to the foot of League One with a 2-0 home defeat against Burton Albion on Saturday.
The Shrews, who sacked manager Paul Hurst earlier this month, have suffered three straight league defeats and have collected just two wins from their opening 15 league games (drawing twice, losing 11).
Sadler made 213 appearances for Shrewsbury as a player but insists his connections to the club will be irrelevant come the first whistle.
"They're having some tough times. I know Paul (Hurst) and Chris (Doig) who've just lost their jobs there really well," he concluded.
"I was gutted that that happened because I've got huge respect for both of them.
"I know the club well. I had some good times there a long time ago it feels know but that'll be irrelevant when the game comes around."
