Karl Hawley missed a golden opportunity to give the hosts the lead in first half when he fired over the bar through on goal.
Instead it was the visitors who grabbed the lead just before the hour mark when Constable stabbed home from close range.
Payne then made it two before Hawley's miserable day was compounded when he had a late penalty saved.
It is the third time in five seasons the Glassboys have been knocked out at this stage of the competiton, while Eastleigh advance to the third round for the first time in their history.
