Paul McCone and Sean Evans netted the second-half goals as the Glassboys' part-timers deservedly defeated their 10-man opponents 2-0.
Gary Hackett's Southern League Premier Division side went ahead on 53 minutes when former Wolves trainee defender McCone headed home from a Sean Geddes cross.
Then ex-Manchester United academy prospect Evans – picked to replace the suspended Aaron Drake – made it 2-0 on 72 minutes by converting a rebound stylishly after Ryan Rowe's shot was saved by goalkeeper Jake Cole.
Argyle had been reduced to 10 men before half-time when defender Paul Bignot was dismissed for kicking out at Geddes.
But the Glassboys dominated throughout with Rowe drawing a fine first-half save from Cole and both Paul Lloyd and Leon Broadhurst hitting the crossbar after the break.
The War Memorial Ground's highest crowd for 37 years – 2,519 – witness the game that was screened live by ESPN.
And the ground will now stage the second-round clash with League One Stevenage.
