The Scholars make the long trip east to face Blue Square Southern division side Cambridge City in the second qualifying round.
Cambridge play at the same level as Blyth Spartans who Chasetown famously beat after a replay to secure a lucrative first round home tie against Oldham two years ago.
And Blakemore is hoping his table-topping Southern League Midlands outfit can do it again.
"It's a massive game for us but we know we're huge underdogs, but just as we were for Blyth and Oldham two years ago, we're looking forward to it," said Blakemore.
"They're a big non-League club and they've got a big history so it will be a good test regarding how far we've come. If we can compete then I'll be pleased.
"I'll just be telling the players not to let themselves down and not to come back with any regrets.
"We've had them watched but we're not overawed and if we can come back to the Midlands with a result, then Chasetown will be back on the map."
Chasetown are again enjoying a professional touch after following their Cup tradition by being booked into a hotel for a pre-match meal.
"We're having a meal like we did before every round two years ago," said the boss.
"The FA Cup will always have a special place in our hearts because it transformed this club then and if we can do half as well again this time then that will be superb."
Blakemore is set to wheel out his big guns after resting all 11 players who helped beat Aylesbury last Saturday for Tuesday's Errea Cup tie against Bedworth.
