It comes the number of spaces at the popular North Walls facility is cut from 45 to nine while a new Morrisons supermarket is being built in Kingsmead.
But 100 new spaces are being created at the Kingsmead site during December. And the rest will be at other locations in the town not currently used for car parking, though the council has not yet revealed where they will be.
Borough council spokesman Will Conaghan said the locations would be revealed shortly.
The access route for vehicles between North Walls and Kingsmead Short Stay has also been closed, however a pedestrian walkway remains in place.
Alternative short stay parking is available on Doxey Road.
Council leader Mike Heenan said it was hoped the new spaces would boost trade in the town centre during the festive period.
Just for Pets and B&M have also been signed up for the Kingsmead park, which will stand next to the £70 million Riverside complex that will feature a new cinema, Marks & Spencer and Primark. Diggers moved onto the clinic site last week and within days it had been flattened.
Planning permission was granted in February 2012 for the superstore along with a petrol station.
Morrisons has taken out a 25-year lease for the 71,000 sq ft food store with 460 car parking spaces.
The redevelopment of the site will also include 14,000 sq ft for other shops which are yet to be confirmed.
