It comes as the trust celebrates its 50th anniversary this year, and a number of celebrations are planned to mark the occasion.
Sue Chance, commercial manager, said: “We’d like to say a big thank you to all those who came to the opening of the shop on Wednesday. We were so pleased to see everyone and are looking forward to seeing our new customers in the coming weeks.
“This is our third charity shop – we also have shops in Leek and Rugeley – and are delighted to be part of Penkridge and how the town has welcomed us.
“The shops help us support our work across Staffordshire, looking after wildlife and wild places in our county.”
The new shop will be open from 9.30am to 4.30pm Monday to Saturday.
