Emergency services closed the road outside the Tesco Express and Esso garage.
Six fire crews were sent to the store in Barnes Hill, Weoley Castle, in the early hours of today.
Police and ambulance services were sent to the scene as firefighters tackled the blaze.
Residents were asked to keep their windows closed as crews fought the fire using a hydraulic platform.
Barnes Hill was closed northbound between Overfield Road and Davena Drive at around 1am by police.
The blaze was put out at around 4am, with electricity being isolated to the store.
It is not yet known what caused the fire, with further investigations set to take place.
West Midlands Fire Service said crews remained at the store into the morning as an investigation into the cause got under way.
The fire started in a petrol station store room and affected 20 per cent of the building, a fire service spokesman said.
