Wolverhampton-based Pc Ben Clay entered a not guilty plea to the charge during a hearing at Wolverhampton Crown Court and will face trial on a date yet to be fixed.
The 39-year-old, who spoke only to enter his plea, was told that the case against him is likely to be transferred to Nottingham Crown Court.
Clay, who was granted conditional bail, is alleged to have committed the offence in June 2014 in the West Midlands area.
West Midlands Police confirmed in December last year that Clay, whose address cannot be published, was arrested after an allegation was made in January 2015.
