Sheila Fox was last seen in Coventry city centre in 1972, aged 16.
At the time, she was living with her parents and is suspected to have been in a relationship with an older man.
Police have found no trace of her has been found since she disappeared decades ago.
It is said to be one of West Midlands Police's longest-running missing person cases.
The force said while it is "entirely possible" she left the area, changed her name and lived a normal life, there are "no traces" of Sheila - who would now be around 68 - in any public record systems.
A police spokesman added: "It is believed Sheila's parents are now deceased, so we are hoping anyone out there who might know what happened to her will get in touch with us and let us know."
Anyone with information about her whereabouts is asked to contact the West Midlands Police Missing Persons Investigation Unit on 0121 626 7003.
